What is the effective inside measurements?

Important part is that it is 7/8" deep, a bit smaller than the size of a regular house key laying flat. This is important because when the key inevitably falls of the shallow hook, the key can barely fit in there with the door of the holder closed. If it is off the hook at the top, it must be angled up a bit before you close the compartment. The other dimensions are 2 1/4" wide and 3 7/8" high. These are all the inside dimensions of the key holder compartment.

Is it 100% waterproof?

I can't speak for it being absolutely 100% waterproof (like dropping it in a bucket or body of water I know it's not waterproof in that sense). The keypad cover will keep out rain and snow, which is important in colder climates where if water penetrates the mechanism and it freezes it could render the lockbox inoperable until it's thawed. Where I have mine mounted on a patio door rain does blow against the patio door, the keypad cover keeps water from getting in the keypad so it serves it's purpose.

Does this fit over the doorknob

Yes this lock box is designed to attach to an existing doorknob. The shackle can be locked or unlocked off of a doorknob when the lock box is opened. Thank you, NLS Products.

Do the numbers/letters have to be entered in sequence or can the correct combination be entered randomly?

Each number/letter can only be pushed once, in any sequence in random, will unlock it. As long as you have pressed the clear button each time.
