The headphones have priority, once unplugged desktop speakers take over.

I use the Fulla 3 between my computer's USB and an outboard power amp. I listen to music, attend Zoom "meetings," and use it for a very convenient volume control. So, while I can't actually answer your specifically, but can say that when I am on Zoom, the headphones would work the same as my speakers do... no feedback from the microphone in my camera. An external microphone, plugged into the Fulla... I am not sure. Amazon's awesome return policy would allow you to return it, no problem, so trying isn't out of the question. I would try it and return it if needed. I love this DAC! It is very well made, solid as a tank, and it worked straight out of the box (Win 10). Hope this helps. Sorry if not.

Do they have enough power for 250 ohm headphones?

Yes, a friend of mine uses dt 990 pros which are 250ohms and they sound great. If you don't have hearing loss, then you should be fine.

80ohms is ez for fulla3. Maximum Power, 16 ohms: 400mW RMS Maximum Power, 32 ohms: 250mW RMS Maximum Power, 50 ohms: 200mW RMS Maximum Power, 300 ohms: 40mW RMS Worry about high impedance headphones like dt990 250ohms. Coz it just sounds as loud as my razer kraken on my mobo's sound card.
