this works with 1/4in tubing right?

No, it's designed for the 8mm tubing that comes with Blumat kits and is the size of the tees that come with each Blumat carrot. We do have fittings though that reduce from this to 1/4."

Can I use this for drilling a hole in a 5 gallon bucket to make my own water reservoir, or do i need another connector to attach to this on the bucket

This is all you should need - you unscrew the left half and it with the washer go inside the bucket, the right side on the right.

Does it come with 1 or 2 adapters?

This is just the single bulk bulk-head fittings, if you buy our buckets which comes pre-drilled, those come with 2.

What size drill bit should be used?

1/2 inch, you could use a hole saw but I would prefer a unibit which can be found in the electrical section. It makes a much cleaner cut than the hole saw.

Does exactly what I expected it to do, air flow with a tight seal, I felt it was a little small at first, but I needed not worry, I used it for air flow though, so, I cannot give advice on flow rates on water with a pump or hydrostatic head.
